Lead-Seeking Approaches (2010)
High quality leads provide the foundation for the discovery of successful clinical development candidates, and therefore the identi?cation of leads is an essential part of drug discovery. The process for the identi?cation of leads generally starts with the screening of a compound collection, either an HTS of a relatively large compound collection (hundreds of thousands to one million plus compounds) or a more focused screen of a smaller set of compounds that have been preselected for the target of interest. Virtual screening methods such as structure-based or pharmacophore-based searches can complement or replace one of the above approaches. Once hits are identi?ed from one or more of these screening methods, they need to be thoroughly characterized in order to con?rm activity and identify areas in need of optimization. Finally, once fully characterized hits are identi?ed, preliminary optimization through synthetic modi?cation is carried out to generate leads. Parallel optimization of all properties, including biological, physicochemical, and ADME is the most ef?cient approach to the identi?cation of leads. Hit characterization is described in the previous chapter. The focus of this chapter is on hit optimization and the identi?- tion of leads. After a general overview of these processes, examples taken from the literature since 2001 will be used to illustrate speci?c points. In 2003 the German Research Foundation established a new priority programme on the subject of "Imaging Measurement Methods for Flow Analysis" (SPP 1147). This research programme was based on the fact that experimental ?ow analysis, in addition to theory and numerics, has always played a predominant part both in ?ow research and in other areas of industrial practice. At the time, however, c- parisons with numerical tools (such as Computational Fluid Dynamics), which were increasingly used in research and practical applications, soon made it clear that there are relatively few experimental procedures which can keep up with state-of-the-art numerical methods in respect of their informative value, e.g. with regard to visu- spatial analysis or the dynamics of ?ow ?elds. The priority programme "Imaging Measurement Methods for Flow Analysis" was to help close this development gap. Hence the project was to focus on the investigation of ef?cient measurement me- ods to analyse complex spatial ?ow ?elds. Speci?c cooperations with computer sciences and especially measurement physics were to advance ?ow measurement techniques to a widely renowned key technology, exceeding the classical ?elds of ?uid mechanics by a long chalk.

A flexagon is a motion structure that has the appearance of a ring of hinged polygons. It can be flexed to display different pairs of faces, usually in cyclic order. Impacts of Point Polluters on Terrestrial Biota: Comparative Analysis of 18 Contaminated Areas (2009)
The adverse consequences of pollution impact on terrestrial ecosystems have been under careful investigation since the beginning of the twentieth century. Several thousand case studies have documented the biotic effects occurring in contaminated areas. However, after more than a century of research, ecologists are still far from understanding the effects of pollution on biota. Only a few generalisations have been made on the basis of extensive monitoring programs and numerous expe- ments with industrial contaminants. The need to reveal general patterns in the responses of terrestrial biota to ind- trial pollution and to identify the sources of variation in these responses became obvious more than a decade ago. At about that time, our team initiated a quanti- tive research synthesis of the biotic effects caused by industrial pollution, based on 1 a meta-analysis of published data. All meta-analyses conducted so far (covering diversity and abundance of soil microfungi, diversity of vascular plants, diversity and abundance of terrestrial arthropods, and plant growth and reproduction) c- sistently showed high heterogeneity in the responses of terrestrial biota to industrial pollution. At the same time, they demonstrated an unexpected shortage of infor- tion suitable for meta-analyses, as well as a considerable influence of methodology of primary studies on the outcome of the research syntheses. To overcome the identified problems, we designed a comparative study, the results of which are reported in this book.
